Well I can think of several locations and ways for character generation: Places like Miami, Chicago, Boston are all places where generally Superheros haven't been completely based well except chicago I think, anyway as for character improvision my two old characters were Russian immigrants and one was former Spetznaz who underwent restricited scientific reaserch that produced a Soldier with skin as tough a wrought iron but it also had... reprocussions for one he accepted this testing despite knowing it was illegal and only survived due to goverment tests coming back negative, the powers were latent and diddn't become active until he moved to America to go live with his arsonist brother and yes I took some of that from GTA IV despite never playing it you may not notice it at first but alot of what we make up tends to have some connection to something we've seen in movies, video games, etc. and if you use too much of that your character goes stale because it's not really your idea and you can't generally plan for them because you don't know they go about doing things. By trying hard not to use unorginal characters and making your own you get much more interesting and diverse characters. Once again these are only theories and may dismissed at your whim or you can read it and gleam some good from it all.
